Prepare for nutrient agar |

Add 5.6 g of nutrient agar into 200 ml of distilled water. Heat the solution until the entire nutrient agar is dissolved. |
Make dye |

Make 1% of Methyl Blue, Eosin, Methyl Green and Malachite Green dyes separately. Mix the solutions by stirring. |
Serial dilution of solution |

Add 28 g of soil into 700 ml of distilled water.
Mix by swirling.
Pipette 1 ml of the soil sample into 99 ml of distilled water in beaker A and stir it. Pipette 1 ml of the diluted soil sample in beaker A into 99 ml of distilled water in beaker B and stir it. |

Transfer soil sample |

Pipette 0.2 ml of soil sample in beaker B into petri plates containing agars with Methyl Blue, Eosin, Methyl Green and Malachite Green respectively. Spread the soil samples evenly around the plates. |
Seal plate |

Seal the plates. |
Keep warm |
|

Turn the plates over and leave them in the oven at 37℃ for 48 hours. Then examine the plates. |
